| 内容記述 | We conducted genetic and phylogenetic analyses of genome segment 2 (Seg-2), which encode serotype-specific structural proteins of the outer capsid, of bluetongue virus (BTV) isolated in Japan from 1985 through 2008. The Japanese strains of BTV were clearly sorted into 6 groups by several genetic characteristics of Seg-2 including segment length, open reading frame length, and 5 '- and 3' - terminal sequences, and they were identified as serotypes 2, 3, 9, 12, 16 and 21 by phylogenetic comparisons with Seg-2 of reference and field strains of 24 serotypes (1 to 24). Thus, our present study revealed that at least 6 serotypes of BTV were isolated in Japan. Our data would be beneficial for understanding BTV epidemiology and for taking better control measures against bluetongue in Japan and its neighboring countries in the Asia-Pacific region. | |||||||||||||||||||||||||||||

| 内容記述 | 本研究では, 1985-2008年に日本国内で分離されたブルータングウイルス (Bluetongue virus:B TV)の遺伝子解析および分子系統樹解析を実施した. 外殻カプシドの構造タンパク質VP2をコードするゲノム分節2の分子系統樹が血清型1-24に完全に一致した分岐を示すことを踏まえて (Maan et al., J. Gen. Virol.88, 621-630 (2007)), 本研究ではそのゲノム分節2を解析の対象とし, BTV日本国内分離株の血清型の同定を試みた. 遺伝子解析の結果, 分節の長さやオープンリーデイングフレームの位置, 5'および3'末端の塩基配列を含む遺伝学的特徴によって, 供試株は6つのグループに明瞭に分類された. さらに, これらの6つのグループは, 分子系統樹解析による血清型1から24の参照株や野外株との比較の結果, それぞれ血清型2, 3, 9, 12, 16および21であることが明らかになった. 本研究の結果により, 国内には少なくとも6つの血清型のBTVが存在することが明らかとなった. 本研究の結果は, 日本および他の近隣アジア諸国におけるBTVの疫学を理解し, より効果的な疾病制御対策を講じる上で有益と考えられる. | |||||||||||||||||||||||||||||
